Sinopse: 

Uma agente do FBI, Olivia Dunham (Anna Torv) se vê obrigada a trabalhar com um cientista famoso, Dr. Walter Bishop (John Noble) que estava internado em um hospital psiquiátrico. Eles têm que desvendar um misterioso caso envolvendo um vírus mortal. Para isso, contam com a ajuda do genial e rebelde Peter Bishop (Joshua Jackson), filho do cientista. Após o episódio piloto, os três continuam a parceria e vão resolver outros casos misteriosos.
